Solarbio ELISA试剂盒采用基于双抗体夹心法的酶联免疫吸附检测技术。将抗小鼠IL-6单克隆抗体包被在酶标板上;分别加入梯度稀释的标准品和预稀释的样本,标准品和样本中的小鼠IL-6会与酶标板上的包被抗体充分结合;洗板后加入生物素化抗小鼠IL-6抗体,该抗体会与板子上包被抗体捕获的标准品和样本中的小鼠IL-6发生特异性结合;洗板后加入辣根过氧化物酶(HRP)标记的链霉亲和素,生物素与链霉亲和素会发生高强度的非共价结合;洗板后加入显色剂底物TMB,若反应孔中样品存在不同浓度的小鼠IL-6,则HRP会使无色TMB变成不同深浅(正相关)的蓝色物质,加入终止液后反应孔会变成黄色;最后,在λmax=450 nm(OD=450 nm)处测定反应孔样品吸光度(OD),样本中的小鼠IL-6浓度与OD成正比,通过绘制标准曲线和四参数拟合软件便可计算出样本中小鼠IL-6的浓度。
背景介绍
白细胞介素-6(IL-6)是由纤维母细胞、单核/巨噬细胞、T淋巴细 胞、B淋巴细胞、上皮细胞、角质细胞、以及多种瘤细胞所产生。IL-1、 TNF-α、PDGF、病毒感染、双链RNA及cAMP等,均可诱导正常细胞 产生白介素6。白介素6能够刺激参与免疫反应的细胞增殖、分化并提 高其功能。IL-6由2条糖蛋白链组成;1条为α链,分子量80KD;另1 条为β链,分子量130KD。α链缺少胞内区,只能以低亲合性与IL-6结 合,所形成的复合物迅即与高亲和性的β链结合,通过β链向细胞内传递 信息。IL-6主要有以下生物特性:诱导B细胞分化;支持浆细胞瘤和骨 髓瘤增生;诱导IL-2和IL-2受体表达;诱导单核细胞分化;诱导CTL; 增强NK细胞活性;诱导急性期反应分子并刺激肝细胞;诱导神经元分 化;诱导肾小球膜细胞生长;诱导角质化细胞生长。
