GLP-1R(Glucagon-Like Peptide-1 Receptor、胰高血糖素样肽-1 受体)属于促胰液素(B1 类)GPCR 家族,主要介导胰高血糖素样肽-1(GLP-1)的生理功能,核心功能为刺激胰岛素分泌、抑制胰高血糖素、延缓胃排空、降低食欲。作为 2 型糖尿病和肥胖治疗的热门靶点,GLP1R 的激活可模拟“肠-脑-胰轴”的天然调控机制,成为代谢性疾病药物研发的焦点。单靶点药物(如 Semaglutide)长期使用可能引发胃肠道副作用(恶心、呕吐),且对脂肪代谢和能量消耗的调控有限。 GCGR(胰高血糖素受体)是一种 B 类 GPCR,最初被描述为与腺苷酸环化酶功能相连的胰高血糖素结合体。GCGR 促进肝糖原分解、脂肪氧化和能量消耗,但过度激活可能导致高血糖。与 GLP-1R 联用可抵消升糖风险,同时增强减重效果。 | |
