Background
Noggin is a secreted homodimeric glycoprotein that functions primarily as an antagonist of bone morphogenetic proteins (BMPs). It regulates BMP activity by binding to specific BMPs—such as BMP4 with high affinity and BMP7 with lower affinity—thereby blocking their interaction with both type I and type II receptors. Structurally, Noggin contains an N-terminal acidic region, a central heparin-binding domain, and a C-terminal cysteine-knot motif. It is expressed in various adult tissues, including the central nervous system, lung, skeletal muscle, and skin. During development, Noggin plays critical roles in neural tube formation, joint morphogenesis, and chondrocyte regulation. Mutations in its cysteine-knot region are linked to skeletal dysplasias. In stem cell biology, Noggin is widely used to maintain pluripotency in human embryonic stem cells and to support neural stem cell maintenance or differentiation into dopaminergic neurons under specific culture conditions.
